Background
The postsynaptic density (PSD) of excitatory synapses is a specialized structure in which an array of synaptic membrane proteins is physically linked and functionally coupled to a specific set of cytoskeletal and/or signaling proteins. The synaptic protein PSD95 binds to and clusters a variety of membrane proteins, including Shaker-type potassium channel subunits and NMDA receptor NR2 subunits, via its first 2 N-terminal PDZ domains. This clustering function implies a direct or indirect interaction with the cytoskeleton. CRIPT (cysteine-rich interactor of PDZ3) binds selectively to the third PDZ (PDZ3) domain of PSD95 via its C terminus. Human CRIPT is a 101-amino acid protein that terminates in a glu-thr-ser-val sequence, consistent with the PDZ-binding tetrapeptide consensus. It shares 99% amino acid sequence identity with rat CRIPT. CRIPT colocalizes with PSD95 in the PSD of excitatory synapses throughout the brain, but is not detected in inhibitory synapses. CRIPT, PSD95, and tubulin may be coimmunoprecipitated from synaptosomal membranes. In heterologous cells, CRIPT caused a redistribution of PSD95 to microtubules. Niethammer et al. (1998) suggested that CRIPT may regulate PSD95 interaction with a tubulin-based cytoskeleton in excitatory synapses.
