Background
Highly conserved coronin family of proteins/actin regulators from WD-repeat superfamily were originally identified in Dictyostelium, wherein, coronin deletions were observed to affect actin-dependent processes, such as phagocytosis, cell motility, and cytokinesis. Coronin-1a (Coro1A) belongs to Type I coronins, almost exclusively expressed in hematopoietic lineages, and plays a key role during T lymphocyte activation, migration, survival, and calcium signaling. Moreover, Coronin-1a has been documented to be dispensable for actin-dependent functions in macrophages. Coro1a-deficient mice have reduced peripheral T cells due to both an impaired survival of naive cells and migrational defects. Both CD4+ and CD8+ T cell subsets are decreased in the periphery of Coro1a-/- mice while thymic cellularity and subpopulations are similar to those observed in wild-type mice. A nonsense mutation in Coro1a has been shown to protect against systemic lupus in a mouse model. Moreover, it has shown that Coro1a is mutated in a thymic egress-deficient mouse strain, but also in a patient with severe combined immunodeficiency (SCID).
