Background
Caspases are a family of cysteine proteases that play key roles in apoptosis. The precursor form of all caspaes is composed of a prodomain, and large and small catalytic subunits. The active forms of caspases are generated by several stimuli including ligand receptor interactions, growth factor deprivation and inhibitors of cellular functions. All known caspases require cleavage adjacent to aspartates to liberate one large and one small subunits, which associate into a2b2 tetramers to form the active enzyme. Caspase 14 also known as Mini ICE (MICE) is a member of the peptidase family C14 that has three potential isoforms. The pro form of Caspase 14 is approximately 29 kD. This caspase may be involved in the death receptor and granzyme B apoptotic pathways. Caspase 14 functions as downstream signal transducer of cell death that can be suppressed by retinoic acid. Caspase 14 becomes activated after granzyme B, Caspase 8, and Caspase 10 cleavage into large p18 and small p10 subunits. This caspase has been shown to associate with the large prodomain caspases, including Caspase 1, Caspase 2, Caspase 4, Caspase 8, and Caspase 10. Its high expression in embryonic tissues and limited expression in adult tissues suggests that it may have a role in ontogenesis.
