NB120-14821 recognizes alpha A crystallin, a 20kDa water-soluble major structural protein and member of the heat shock protein family, designated HSPB4, which acts as a molecular chaperone and is believed to play a role in the maintenance of ocular lens transparency/refractive index. Besides the involvement of both alpha A and alpha B crystallin in the prevention of protein precipitation and cataract formation in the eye, alpha crystallins have also been demonstrated in other tissues including the heart, brain, kidney, spleen and thymus. |

Background
Alpha crystallins are major proteins of the ocular lens and are thought to play a role in maintaining lens transparency. They are composed of two gene products alpha A and alpha B, for acidic and basic, respectively. Alpha-crystallins can be induced by heat shock and are members of the small heat shock protein (sHSP) family. They act as molecular chaperones and hold unfolded or misfolded proteins in large, water-soluble low molecular weight aggregates. These heterogeneous aggregates consist of 30-40 subunits of alpha crystallins, and the alpha A and alpha B subunits have a 3:1 ratio, respectively. Two additional functions of alpha crystallins are an autokinase activity and the participation in the intracellular architecture.
