Background

Fibrinogen is the main protein of the blood coagulation system. It is a large protein and it consists of two identical subunits that contain three polypeptide chains: alpha, beta and gamma. All chains are connected with each other by a number of disulfide bonds. Fibrinopeptides A and B are released by thrombin from the N terminal parts of alpha and beta chains, respectively. In this way fibrinogen is converted into fibrin, which by means of polymerisation forms a fibrin clot. Fibrinogen clotting underlies pathogenesis of MI, thromboembolism and thromboses of arteries and veins, since fibrin is the main substrate for thrombus formation. Fibrinogen activation is also involved in pathogenesis of inflammation, tumor growth and many other diseases.

  1. Have you tested this product to determine reactivity to fibrinogen and fibrin? Can you email me the results? Before ordering we need proof this antibody reacts only to fibrinogen and not to fibrin.
    • I do not see any specific notes or testing data on file from the lab indicating direct testing for reactivity to fibrin.
  2. So this is a product with no negative control? Hasn't someone at least tested it on tissue such as skin from someone with active angiogenesis to make sure it doesn't bind clots?
    • I heard back from the lab this morning and they had indicated that unfortunately we can't say for sure that our anti-fibrinopeptide A antibody will not detect fibrin. It was believed for a long time that fibrinopeptide A is present only in fibrinogen and fibrin did not contain it when this antibody was synthesized. The latest data shows however, that fibrin contains a significant amount of uncleaved fibrinopeptide A that is whole uncleaved fibrinogen molecules are incorporated into fibrin. As a result we do not have any direct testing data on file that we can send to show with certainty that it will or will not detect fibrin.
