Stimulation with IL-1 beta resulted in production of COX-2, MMP-9, MMP-13 and caspase-3 cleavage. Pre-treatment with a combination of both IGF-1 or/and PDGF-bb downregulated COX-2, MMP-9, MMP-13 and cleaved caspase-3. Curcumin inhibits IL-1 beta activity, enabling co-culture induced chondrogenesis in MSCs. A: Fourteen days high-density culture. Untreated MSC cultures became apoptotic (a). Immunohistochemistry-Paraffin: SREBP1 Antibody [NB100-2215] - nalysis of FFPE tissue section of mouse liver with SREBP1 antibody at 1:200. The antibody generated an expected cytoplasmic-nuclear immunostaining of SREBP1 protein in the hepatocytes.

The PPAR alpha activator, Irbe, reduces SREBP1 expression and induces the expression and co-localization of PPAR alpha and p21/p27 in Ishikawa cells. Immunocytochemistry/Immunofluorescence: SREBP2 Antibody [NB100-74543] - Analysis of SREBP2 (green) showing staining in the cytoplasm of L6 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with a SREBP2 polyclonal antibody in 3% BSA-PBS at a dilution of 1:100 and incubated overnight at 4C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight-conjugated secondary antibody in PBS at room temperature in the dark. F-actin (red) was stained with a flourescent red phalloidin and nuclei (blue) were stained with Hoechst or DAPI.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. NBP3-32977 at 1/2,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG [HRP] Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.
