![Immunocytochemistry/ Immunofluorescence: Calbindin D-28K Antibody [NBP2-50028] - Myh14 localizes to apical membranes of Atp6v1b1‐positive intercalated cells in the distal tubule. Adult kidney sections were stained for calbindin‐D28K, Atp6v1b1, Aqp2, & Myh14 to identify intercalated cells within the distal & connecting tubule segments as well as to confirm Myh14 localization to the intercalated cell type. Cells with positive staining for Atp6v1b1 (red) can be found in tubule segments positive for calbindin‐D28K only (A–D) as well as in tubules positive for both calbindin‐D28K & Aqp2 (E–H); in both cases Atp6v1b1 staining is observed along the basolateral membrane indicating beta ‐ intercalated cell type (arrowhead, D&H). We also observed Atp6v1b1 staining in majority of the tubules along the apical membrane indicating the alpha ‐ intercalated cell type (arrowhead, I–L). Myh14 localized to the apical membrane of the intercalated cell type in the distal tubule (arrowhead, M–P). ![Western Blot: Calnexin Antibody - BSA Free [NB100-1974] - Immunoblot analysis of livers from control & L-Chrebp−/− mice subjected to fasting & refeeding with a high-sucrose diet. Littermate control & L-Chrebp−/− mice (same as those described in supplemental Tables S2A & S2B) were subjected to fasting & refeeding. The nonfasted (N) groups were fed chow diet ad libitum. The fasted (F) group was fasted 12 h, & the refed (R) group was fasted for 12 h & then refed with 60% (w/w) high-sucrose diet for 12 h prior to study. Liver whole-cell lysates & membrane fractions were prepared individually, & equal amounts of protein from each mouse of the same group (four per group) were pooled. Aliquots (40 μg for whole-cell lysates & 30 μg for membrane fractions) of the pooled protein were subjected to SDS-PAGE & immunoblot analysis. Immunoblot analysis of Insig-1 & Insig-2 were carried out using membrane fractions. Whole-cell lysates were used to detect other proteins. ![Western Blot: Vimentin Antibody (2D1) [NBP1-92687] - High glucose (HG) concentrations induced epithelial-to-mesenchymal transition protein expression & enhanced migration activity in colorectal cancer (CRC) cells. SW480 (low metastatic potential) & SW620 (high metastatic potential) cells were cultured in different concentrations of glucose (normal: NG; HG; & osmotic control: NG + l-glucose). (A) Morphological change occurred from epithelial to mesenchymal type in the HG-concentration group. (B) HG concentration caused downregulation of E-cadherin & upregulation of N-cadherin, beta CTN, & vimentin, but c-myc was unchanged, as detected using Western blotting. beta -actin was evaluated as an internal control. (C,D) Wound healing assay showed that HG concentration promoted cell motility in SW480 & SW620 CRC cells after 48 & 72 h of culture, compared with the NG & NG + l-glucose groups. (E) In a Transwell migration assay, 3.5 × 105 SW480 & SW620 CRC cells were plated onto a 24-well plate & cultured in NG & HG-concentration medium for 96 h. HG concentration promoted cell motility in SW480 & SW620 cells. NG + l-glucose cells were evaluated as ostomic controls. (F) These data show that HG concentration caused upregulation of p-IGF1R in CRC. In addition, HG concentration promoted IGF1R downstream signaling, including p-Src & p-ERK; these proteins were increased when CRC cells were cultured in HG-concentration medium. Levels of beta -actin were evaluated as loading controls.
