The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Nuclear staining was observed. ![Immunocytochemistry/Immunofluorescence: TARDBP Antibody (3H8) [NBP1-92695] - NBP1-92695 was used to stain a section of formalin fixed adult rat brain, specifically the hippocampus. Hippocampal neuron nuclei are stained strongly. Chicken antibody to GFAP (NBP1-05198, green) shows the processes of astrocytic glial cells. Nuclei of all cells are revealed with DAPI DNA stain (blue). The TARDBP (3H8) antibody stains neuronal nuclei strongly and the nuclei of some non-neuronal cells much more weakly. ![Immunohistochemistry-Paraffin: TDP-43/TARDBP Antibody (JM51-10) [NBP2-80137] - Immunohistochemical analysis of paraffin-embedded human spleen tissue using anti-TDP-43/TARDBP antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes. The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (1/50) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen.
