Obacunone prevents SSR-induced p53 and p21 gene and protein expression modulations in a FT skin model. FT skin models were pretreated for 24 h with obacunone (12.5 uM, 25 uM, and 50 uM) and subjected to 18 J/cm2 of SSR. (A) 24 h after irradiation, p21 and p53 mRNA levels were measured by real-time PCR. Data are expressed as 2− delta delta Ct. (B) p21 and p53 levels were analyzed by Western blotting. Quantification was performed by densitometry and normalized to beta -actin. Data are expressed as 2− delta delta Ct. All data are expressed as a scatter plot with mean +/- standard error of the mean (SEM) of at least two independent experiments (n = 3). rhIL-11 and soluble rhIL-11R alpha promotes time-dependent proliferation and senescence in human pulmonary artery endothelial cells (HPAEC) and smooth muscle cells (HPASMC). A HPAECs or B HPASMCs were isolated from control donor subjects and stimulated with rhIL-11 5 ng/ml, rhIL-11R alpha 10 ng/ml or its combination at indicated times. Experiments were done between passages 2–3. Cell proliferation was measured by the BrDU kit at 24 h, 48 h, 72 h and 96 h. Cell senescence was measured after 72 h of cell stimulation using beta -galactosidase histology and P21 expression. Results were expressed as % senescence ( beta -galactosidase blue positive cells) relative to the total number of cells in each field. P21 expression was measured by quantitative PCR (qPCR) as 2− delta Ct and western blot. Blue box: in WT cells, XRCC1 protein complexes limit PARP1 engagement and activity during BER by promoting efficient hand-off of SSB intermediates to POL beta and LIG3, preventing PARP1 from impeding repair. Orange box: in XRCC1−/− cells, the absence of XRCC1 protein complexes results in excessive cycles of PARP1 association/activation at SSB intermediates, which impedes access by other BER enzymes and blocks their repair, resulting in SSB accumulation. If this scenario is sufficiently prolonged, such as at high levels of base damage, then this increased PARP1 engagement leads progressively to NAD+ depletion, declining PARP1 auto-ribosylation and dissociation, and accumulation of PARP1 in chromatin. PARP1 trapping in this scenario thus reflects both increased PARP1 association at SSB intermediates and subsequently decreased PARP1 dissociation, both of which impede BER in a manner reminiscent of chemical PARP inhibitors (pink box shown for comparison). Immunocytochemistry/Immunofluorescence: 53BP1 Antibody (1285C) [NBP2-54753] - Neuro2a cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with 53BP1 Antibody at 5 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) (Catalog #NB100-690) was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Flow (Intracellular): 53BP1 Antibody (1285C) [NBP2-54753] - An intracellular stain was performed on HeLa Cells with 53BP1 (1285C) antibody (Catalog #NBP2-54753) (blue) and a matched isotype control MAB1050 (orange). Cells were fixed with 4% paraformaldehyde, following fixation, cells were permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by rabbit IgG APC-conjugated secondary antibody (F0111, R&D Systems).

Immunocytochemistry/Immunofluorescence: 53BP1 Antibody (1285C) [NBP2-54753] - NIH3T3 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-53BP1 Antibody [1285C] NBP2-54753 at 2 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:1000 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Flow Cytometry: 53BP1 Antibody (1285C) [NBP2-54753] - An intracellular stain was performed on Ntera2 cells with 53BP1 Antibody [1285C] NBP2-54753 (blue) and a matched isotype control NBP2-24891 (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1.0 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (SA5-10033, Thermo Fisher).

Cells were counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunoprecipitation: TAF15 Antibody [NB100-567] - Detection of human TAFII68 on HeLa whole cell lysate using NB100-567. TAFII68 was IPed with rabbit anti-TAFII68 antibodies NB100-566, NB100-567, NB100-568, and another using each at 0.3 ug/mg lysate.

ALS proteins associate with the RNAP II/U1 snRNP machinery in an RNA-independent manner. IPs were carried out from RNase A-treated or untreated nuclear extract using an RNAP II or a negative control antibody (EIF4A3) followed by westerns with the indicated antibodies.

Western Blot: PNK Antibody [NBP1-87257] - Lysine 138, arginine 139 & arginine 141 are important for efficient DNA repair.(A) PNKP depletion by 3’-UTR siRNA PNKP was confirmed by western blot analysis. KAP1 antibody was used for loading control. (B) DNA DSB repair efficiency was analyzed by immunofluorescence using gamma -H2AX antibody after IR exposure in GFP-PNKP & 3’-UTR siRNA PNKP-transfected U2OS cells. Graph represented percentage of gamma -H2AX positive cells with 10 ≧ gamma -H2AX foci. At least 200 cells were counted & experiments were performed 3 times independently. (C) DNA SSB repair efficiency was analyzed by immunofluorescence using PAN-ADP-ribose binding reagents at 30 mins after IR exposure in GFP-PNKP & 3’-UTR siRNA PNKP-transfected U2OS cells treated with 10μM of poly (ADP-ribose) glycohydrolase inhibitor (PARGi) for 30 mins prior to IR exposure. (D) Graph represented the intensity of ADP ribose. At least 100 cells were counted & experiments were performed 3 times independently.
