Background
Interferon-alpha 13 (IFN alpha -13), also known as IFN alpha -1/13 and
LeIF D, is one of 14 subtypes within the IFN-alpha family (1). Interferons are a
family of cytokines with potent antiviral, antiproliferative and immunomodulatory
properties, classified based on their binding specificity to cell surface
receptors (1). The members of the IFN-alpha family are distinct but closely related
proteins sharing approximately 80% amino acid (aa) sequence identity and a globular
structure composed of five alpha-helices (1-3). The mature extracellular
domain (ECD) of mouse IFN alpha -13 is shares 64% aa sequence identity with human
IFN alpha -13. IFN-alpha family members signal through a common set of cell surface
receptor complex composed of IFN alpha -R2 and IFN alpha -R1 subunits (2). Individual IFN-alpha
subtypes are known to display unique efficacies to viral protection, and IFN alpha -13
displays antiviral activity protective against Theiler's virus, Mengo virus,
VSV (4). IFN alpha -13 reproducibly appears to be up-regulated in response to TMEV
infection (5). IFN alpha -13 also protects L929 cells against EMCV as tested in our
assays.
