Alternate Names for NSD3 Antibody
Background
NSD family of SET domain-containing histone methyltransferases includes NSD1, NSD2 (also known as WHSC1, Wolf-Hirschhorn syndrome candidate 1 or MMSET, multiple myeloma SET), and NSD3 (also known as WHSC1L1, WHSC1 like 1). All the three NSD proteins have been directly linked to multiple human diseases and each NSD family member behaves as an oncogene. HSD3 is a istone methyltransferase that preferentially methylates 'Lys-4' and 'Lys-27' of histone H3. H3 'Lys-4' methylation represents a specific tag for epigenetic transcriptional activation, while 'Lys-27' is a mark for transcriptional repression. Translocations resulting in NUP98 fusion to NSD1 and NSD3 leads to the development of acute myeloid leukemia (AML) and overexpression of NSD3 via gene amplification occurs in several breast cancer cell lines as well as in non small cell lung carcinomas (NSCLC).

I have a customer with several questions about the anti-WHSC1L1 antibody (catalog # NBP1-04942). They would like to know the following: Has this antibody been shown to detect the other three isoforms of this protein as shown on Swiss-Prot for Q9BZ95? They are expecting that it should detect the other isoforms because of the location of the immunogenic peptide. What was the exposure time for the Northern blot image shown on the datasheet? Has the antibody been tested in Western blot with any other cell lines or tissue extracts? May I just inquire did you try any additional cell lines or tissue extracts besides HeLa nuclear extracts? I agree with you that isoforms 1, 2 and 4 are very similar in size. Unfortunately, the customer specific asked mentioned isoform 3 which is missing amino acids 646-1437 and should therefore be much smaller. Might you have any testing data that shows detection of isoform 3? I admit I know nothing about which tissue types express which isoform other than what I see on Swiss-Prot.
- The immunogen for NBP1-04942 is present in all isoforms of NSD3 so it should detect all of them. Most are very similar in size, so it may not be possible to distinguish them at such high molecular weights. For the Western blot, we do a variety of exposure times so I am not sure as to which one worked best for this particular image. It looks like we only tested this in HeLa, sorry! It also looks like a few of our test bleeds did detect isoform 3 at around 75 kDa but that wasn't the one we chose for production. As far as tissue specificity, I'm with you that I just have information from Swiss-Prot and the Human Protein Atlas which looks like this is pretty ubiquitous (https://www.proteinatlas.org/ENSG00000147548-NSD3).
