Background
Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. The encoded enzyme is regulated by steroid hormones. In tissue culture, the enzyme has been found to generate amyloidogenic fragments from the amyloid precursor protein, suggesting a potential for involvement in Alzheimer's disease. Multiple alternatively spliced transcript variants that encode different isoforms have been identified for this gene. [provided by RefSeq]

I purchased Human Kallikrein 6 Recombinant Protein (Cat # H00005653-P01) several months ago to use as a standard for an ELISA procedure. The lot number of the material that we received is 1000316. The Technical Data Sheet that accompanied the protein states that This protein is not active and should not be used for experiments requiring activity. My questions: Does this product contain any enzymatic activity? If the protein is not active, how do you assess activity? By enzyme activity assay? If so, can you provide the data indicating no activity?
- This recombinant protein actually happens to be one that we distribute for a Taiwanese company called Abnova. We do not perform any testing in house. The reason that the datasheet indicates that this protein is not active is because of the way Abnova synthesizes their recombinant proteins. They use a cell free wheat germ system that is unique to them. With our experience this system does not produce the same post translational modifications and folding that proteins would undergo in an endogenous cell thus rendering them inactive.
