Background

GPR120 is a member of the rhodopsin family of G protein-coupled receptors (GPRs), which was found in 2003 by Fredriksson et al. GPR120 coded by 377 a.a with 7 transmembrane domain, and ERM motif, hightly expressed in stomach, lung, colon, rectum and ileum. GPR120 regulated calcium mobilization, MAP kinase pathway.

  1. I am interested in one of your antibodies, The cat number is NBP1-70975. It said the Ab can be used for IF, can I used for FACS?
    • We haven't tested it for FACS, therefore we can't guarantee it will work. But, if you want to purchase it to test in FACS, you will qualify for our Innovators Reward Program. Should you decide to use one of our products in an application or species for which it has not been tested.
  2. It is stated in the datasheet that this antibody has been raised against the internal peptide, would you please provide the sequence of the peptide? I would like to know if this peptide is against the cytoplasmic domain or the membranous domain.
    • The amino acid range of this immunogen is aa 232-282 of human omega-3 fatty acid receptor 1 with Q5NUL3 (https://www.uniprot.org/uniprotkb/Q5NUL3/entry).This antibody binds to the third cytoplasmic domain of human omega-3 fatty acid receptor 1.
  3. I noticed that this Ab can be used for IF, how did you fix and permerblize the cells? And can I use it for mouse GPR120?
    • NBP1-70975 has been tested and validated to work in mouse samples. Here is our recommendation for IF fixation and permeabolization, although you may need to optimize this based on your samples: 1. Pull off culture medium and add 10% formalin to each well. Let sit at room temperature for 10 minutes. 2. Take off the formalin and permeabilize cells as follows, depending on protein type: a. For detection of either nuclear or mitochondrial protein, add 1 ml TBS with 0.5% Triton X-100. Incubate for 10 minutes at room temperature. b.For detection of a cytoplasmic or membrane protein, add 1 ml PBS/ with 0.5% Tween-20 for 10 minutes at room temperature.
