Background

DEPTOR, a component of the mTORC1 and mTORC2 complexes, is an inhibitor of mTOR. DEPTOR, along with mTOR, is one of five subunits of the rapamycin-sensitive mTORC1 complex or rapamycin-insensitive mTORC2 complex. mTOR itself is a PI3-kinase which plays a key role in maintenance of cellular homeostasis in response to microenvironments induced by growth factors, cell stress or nutrient conditions (1-3). While DEPTOR inhibits mTOR, in turn DEPTOR is repressed by mTORC1 and mTORC2. Thus control of mTOR, DEPTOR and related mTORC1 or mTORC2 pathways have become an important target for controlling cellular growth, or aberrant cellular growth in cancers, for example. In some multiple myelomas, unlike many cancers where DEPTOR expression is low, DEPTOR is overexpressed and PI3k and AkT high activities maintain cell survival (4). Interestingly the red wine polyphenol Resveratrol, a highly acclaimed treatment for cancer or cardiovascular disease management also appears to enhance mTOR and DEPTOR interaction, leading to mTOR inhibition (5). No surprise that many swear by the positive and beneficial effects of imbibing a glass of red wine with some frequency! Antibody to DEPTOR helps to clarify its expression and role in modulating mTOR driven growth of cancer cells for example.
