Immunocytochemistry/Immunofluorescence: CD44 Antibody (8E2F3) [NBP1-47386] - HeLa cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-CD44 Antibody [8E2F3] conjugated to DyLight 550 (NBP1-47386R) at 5 ug/ml for 1 hour at room temperature. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Western Blot: CD44 Antibody (8E2F3) - BSA Free [NBP1-47386] - Generation and characterization of immortalized primary keratinocytes (IPKs) from K14Cre.Ptpn2w/w and K14Cre.Ptpn2fl/fl mice. Western blot analysis of pSTAT3, STAT3, and CD44 (NBP1-47386) in primary keratinocytes and IPKs from both genotypes. The A431 cells were used as positive controls. Ability of the antibodies to capture CD44 antigen was first assessed by comparing the level of CD44 antigen from the starting material (SM) to its level remaining in the unbound fractions (UB). Anti-CD44 antigen at 1/2000 was used for each immunoblot. Immunoprecipitate for CD44 Antibody (NBP1-47386) that showed depleted CD44 antigen in the UB can be seen. CD44 (8E2F3) was detected in immersion fixed A431 human skin carcinoma cell line using Mouse anti- CD44 (8E2F3) Protein-G purified Monoclonal Antibody conjugated to Biotin (Catalog # NBP1-47386B) at 2 µg/mL overnight at 4C. Cells were stained using Streptavidin conjugated to DyLight 550 (red) and counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

U-251 MG human glioblastoma cell line was stained with Mouse anti-CD44 (8E2F3) Protein-G purified Monoclonal Antibody conjugated to Alexa Fluor® 647 (Catalog # NBP1-47386AF647, blue histogram) or matched control antibody (orange histogram).

Immunohistochemistry-Paraffin: HIF-1 alpha Antibody [NB100-654] - IHC analysis of a formalin-fixed paraffin-embedded tissue section of human endometrium carcinoma AN3CA cell line based xenograft using rabbit polyclonal HIF-1 alpha antibody NB100-654 at 1:300 dilution. The signal was developed using HRP-labelled secondary antibody and DAB reagent, and the section was further counterstained using hematoxylin. Simple Western: HIF-1 alpha Antibody [NB100-654] - Simple Western lane view shows a specific band for HIF-1 alpha in 0.5 mg/ml of Hypoxic HeLa lysate. This experiment was performed under reducing conditions using the 12-230 kDa separation system.

Gel Super Shift Assays: HIF-1 alpha Antibody [NB100-654] - Mouse HIF-1 alpha+/+ and HIF-1 alpha+/- cells were exposed to hypoxia or 125 uM CoCl2 for 6 h, nuclear extracts were then prepared and EMSA carried out. HIF-1 isolated from hypoxia or CoCl2-treated cells bound to the wild-type probe (lanes #1-6) but not to the mutant probe (lanes #7-12). HIF-1/DNA complex was detected in hypoxia (H)-or CoCl2 (Co)-treated cells (lanes #2, 3, 5, 6) but not in control (C) cells (lanes #1, 4). More complex (darker band) was seen in hypoxia-or CoCl2-treated HIF-1 alpha+/+ cells (lanes #2, 3) than that in hypoxia-or CoCl2-treated HIF-1 alpha+/- cells (lanes #5, 6). Immunohistochemistry-Paraffin: TfR (Transferrin R) Antibody (OX26) [NB200-585] - Analysis of FFPE mouse spleen using TfR (OX26) antibody at 1:50 on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Plasma membrane staining was observed. Staining was performed by Histowiz.

Western Blot: RelA/NFkB p65 Antibody (112A1021) [NB100-56712] - Lysates of HeLa human cervical epithelial carcinoma cell line and Daudi human Burkitt's lymphoma cell line. PVDF membrane was probed with 1 ug/mL mouse anti-RelA/NFkB p65 monoclonal (NB100-56712, Novus Biologicals), followed by 1:2000 dilution of the appropriate HRP-conjugated secondary antibody, donkey anti-mouse IgG (HAF018).

Immunohistochemistry-Paraffin: RelA/NFkB p65 Antibody (112A1021) [NB100-56712] - Ovarian cystadenocarcinoma probed with p65 antibody at 5 ug/mL. Human tissue TMA was used for this test. Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10 mM sodium citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min.

Flow Cytometry: RelA/NFkB p65 Antibody (112A1021) [NB100-56712] - Intracellular staining of 293 HEK cells using 0.5 ug of p65 antibody. Green histogram represents the isotype control (p65) antibody. This NFkB p65 antibody was used for this test with an anti-mouse IgG PE conjugated secondary antibody.
