ZMPSTE24 (zinc metalloproteinase Ste24 homolog) is a zinc metalloprotease that catalyzes the removal of C-terminal three residues of farnesylated proteins and is essential for maturation of lamin A (a nuclear scaffold protein). ZMPSTE24 belongs to peptidase M48A family and is a membrane-associated enzyme which contains 7-transmembrane segments localized to ER membrane as well as inner nuclear membrane. ZMPSTE24 contains a consensus zinc metalloprotease motif, consisting of a HEXXH catalytic site, located in a loop predicted to reside in the cytosol/nucleoplasm and farnesylated substrates such as prelamin A are accessible to the HEXXH domain for proteolytic cleavage. Lamin A, together with lamin C and B-type lamins, form the meshwork that underlies the nuclear envelope, and provide structure/shape to the nucleus along with playing many critical roles in nuclear function such as binding of heterochromatin, regulation of transcription factors, positioning of nuclear pore complexes and facilitating response to DNA damage. Defects in ZMPSTE24 have been linked to mandibuloacral dysplasia with type B lipodystrophy (MADB) and lethal tight skin contracture syndrome (LTSCS).

Images were captured and analyzed using Olympus Fluoview Confocal microscope (Olympus, Center Valley, PA)
Slides are treated with cold methanol at -20C for 10 min.
Washed with 1X PBS for 3 times, each time for 5 min.
Cells were cultured in the Lab-Tek II Chamber Slide w/Cover CC2 Glass Slide Sterile, 4-well (Thermo Fisher Scientific, Catalog #154917). After indicated treatment, cells were fixed and permeabilized using cold methanol for 10 min at -20°C. Then the slides were washed with 1X PBS for 10 min and blocked with 1XPBS containing 5% Normal Goat Serum for 1 hr. The slides were incubated diluted anti-ZMP antibody (1:100) at 4°C for 12hr.After 3X washings (5 min each wash) with 1X PBS, cells were incubated with corresponding Alexa Fluor conjugated secondary antibodies (Life Technologies) for 1 hr at room temperature. The slides were washed 3 times (5min each time) with 1X PBS and counter stained with 300nM DAPI for 1 min, followed by 1X PBS washing for 1 min. After air drying, slides were sealed with Gold Seal Cover Glass (Electron Microscopy Sciences, Catalog #3223) using Fluorogel (Electron Microscopy Sciences, Catalog #17985-10). Images were captured and analyzed using Olympus Fluoview Confocal microscope (Olympus, Center Valley, PA).
