Background
Human Inactive rhomboid protein 2 (RHBDF2 or IRHOM2)
is a highly conserved seven-transmembrane Rhomboid protease-like protein which
is inactive, it has no protease activity, but has been implicated in the
secretion of several ligands of the epidermal growth factor receptor (EGFR),
probably by regulating trafficking and stability of key protein membrane like
ADAM17 and STING. The RHBDF2 protein plays an important role in the secretion
of tumor necrosis factor alpha and has been implicated in familial esophageal
cancer. Gain-of-function (GOF) mutations in the RHBDF2 gene constitutively
activate EGFR signaling to cause tylosis with esophageal cancer syndrome.
