Background
Butyrophilin-like
9 (BTNL9) is a member of the BTN/MOG
Ig-superfamily and functions as a negative regulator of immune cell activation
(1). Mouse BTNL9 is a 536 amino acid (aa) type I transmembrane
glycoprotein that contains a 35 aa signal sequence, a 222 aa extracellular
domain (ECD), a 21 aa transmembrane region and a 258 aa cytoplasmic segment. The ECD features two V-type Ig-like domains and three
glycosylation sites (2-4). The ECD of mouse BTNL9 shares ~67% and 91% aa sequence
identity with the ECD of human and rat BTNL9, respectively. BTNL9 is expressed in a variety of tissues
in humans and mice. Among isolated immune cell types, B cells show the
strongest expression (2, 5). Recombinant BTNL9–Fc has been shown to bind to
many immune cells including T cells, B cells, macrophages and dendritic cells
(6). With this profile of binding, it
will not be surprising if BTNL9 joined the panel of butyrophilins with negative
regulatory effects on immune cells (5). BTNL9 has been reported to inhibit CD8
+
T Cell proliferation (5).
