Background

p18 INK4c is a specific inhibitor of cdk4 / cdk6 (interacts strongly with CDK6 and weakly with CDK4), and is expressed during G1 to S transition in the eukaryotic cell division cycle. It is maximally induced as cells enter S phase. Highest expression levels found in skeletal muscle. Also found in pancreas and heart.

PVDF Membrane was probed with 1 µg/mL of Mouse Anti-Human p15INK4b/CDKN2B Monoclonal Antibody (Catalog # MAB6798) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for p15INK4b/CDKN2B at approximately 15 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
p15INK4b/CDKN2B was detected in immersion fixed HeLa human cervical epithelial carcinoma cell line using Mouse Anti-Human p15INK4b/CDKN2B Monoclonal Antibody (Catalog # MAB6798) at 3 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. PVDF membrane was probed with 0.5 µg/mL Human Phospho-p27/Kip1 (T157) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1555) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band for Phospho-p27/Kip1 (T157) was detected at approximately 27 kDa (as indicated). The phospho-specificity of this antibody was supported by decreased labeling following treatment with 600 U lambda -phosphatase ( lambda -PPase) for 1 hour. This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counterstained with hematoxylin (blue).
