Use in WB reported in scientific literature (PMID:33850935) IHC: NBP1-52375 was validated for use in IHC on a panel of 21 IHC-P (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity.

The nicotinic acetylcholine receptors (nAChRs) are members of a superfamily of ligand-gated ion channels that mediatefast signal transmission at synapses. The nAChRs are thought to be hetero-pentamers composed of homologous subunits.The proposed structure for each subunit is a conserved N-terminal extracellular domain followed by three conservedtransmembrane domains, a variable cytoplasmic loop, a fourth conserved transmembrane domain, and a short C-terminalextracellular region. The protein encoded by this gene forms a homo-oligomeric channel, displays marked permeabilityto calcium ions and is a major component of brain nicotinic receptors that are blocked by, and highly sensitive to,alpha-bungarotoxin. Once this receptor binds acetylcholine, it undergoes an extensive change in conformation thataffects all subunits and leads to opening of an ion-conducting channel across the plasma membrane. This gene islocated in a region identified as a major susceptibility locus for juvenile myoclonic epilepsy and a chromosomallocation involved in the genetic transmission of schizophrenia. Would you please tell me whether your antibody Nicotinic Acetylcholine R alpha 7/CHRNA7 Antibody (NBP1-52375) is suitable for detecting rat tissue which is PFA perfusion fixed frozen sections?
We have not tested this product on frozen sections yet but since it worked well in IHC-P, there is a great chance that it will work on IHC-Fr. Technically speaking, IHC-P protocol poses the target proteins/antigens to more harsh conditions/reagents compared to IHC-Fr, ICC-IF and FLOW protocols. And most of antibodies which work for IHC-P application works for IHC-Fr, ICC-IF and FLOW also.
