Background
HVCN1 (hydrogen voltage-gated channel 1), a voltage gated proton channel protein that is highly expressed in immune tissues, mediates the proton conductances required by phagocytic leukocytes for the oxidative burst that underlies microbial killing. HVCN1 moderates the voltage-dependent proton permeability of excitable membranes by allowing the flow of protons in accordance to their electrochemical gradient. HVCN1 is sensitive to zinc ions, and can be inhibited by them. HVCN1 may represent a new therapeutic target for B cell malignancies on continued signaling via the B cell antigen receptor.

-
Could this product be used for Immunofluorescence on cultured cells from mice?
- In our lab, we have not tested this antibody for its ICC-IF application yet and therefore, we have no information weather it will work in ICC-IF application or not. We can not guarantee its ICC-IF application, however, you may optimize the best suitable conditions for the staining from your own testing, and we would be more than happy to provide you our troubleshooting/technical feedback if you come up with any problems in generating your desired staining. For your information, our lab's general protocols for ICC-IF application as well as the associated troubleshooting suggestions may be found at the following weblinks: Immunocytochemistry Protocol and Immunocytochemistry Troubleshooting. If you would be interested in testing this novel application, please take a look at our Innovator's Reward program. Moreover, I would highly suggest you to include our HVCN1 Peptide # NBP1-76278PEP into your ICC-IF assay for HVCN1 detection. This peptide may be used for executing a blocking experiment to confirm your staining findings. The Peptide Competition Protocol may be found at Peptide Competition Protocol.

What is the expected band size of NBP1-76278?
- The canonical isoform of HVCN1 protein is 273 amino acids in length and its expected molecular weight is 31.7 kD. Our HVCN1 Antibody (NBP1-76278) has been raised against 17 amino acids near the carboxy terminus of human HVCN1 and this antibody detected a single specific band just above the 30kD position. We do sell a blocking peptide specific against this antibody - HVCN1 Peptide (NBP1-76278PEP). This peptide may be used for conducting the peptide block assays for proving the specificity of the immunodetection and a general protocol for such assays may be found at peptide competition protocol.
