Background

Complement Component 3a receptor (C3a R) is an ~90 kDa G-protein coupled 7-transmembrane sulfated glycoprotein expressed on myeloid peripheral blood leukocytes, and on activated lymphocytes, endothelial and internal organ epithelial cells. Engagement of C3a R by C3a activates mast cells and neutrophils, triggering degranulation in airways during asthmatic allergen challenges. It assists in Th2-type inflammatory reactions and stimulates leukocyte chemotaxis. Human C3a R extracellular regions share 50% and 42% amino acid identity with mouse and rat C3a R, respectively.

Flow (Intracellular): Complement C3 Antibody (11H9) [NB200-540] - An intracellular stain was performed on RAW 246.7 cells with Complement C3 (11H9-3-2) antibody NB200-540 (blue) and a matched isotype control NBP2-31382 (orange). Cells were either treated with 3uM Monensin for 3 hours to block the secretion of Complement C3 (B) or grown in normal media (A). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2 ug/mL for 30 minutes at room temperature, followed by mouse F(ab)2 IgG (H+L) PE-conjugated secondary antibody (F0102B, R&D Systems).
Immunocytochemistry/Immunofluorescence: Complement C3 Antibody (11H9) [NB200-540] - C3 protein fragments deposited on kidney cells of MPL-lpr mouse. Staining with antibody 11H9. Glomerular staining pattern. Fixation in 4% paraformaldehyde in PBS pH 7.4. Vibratome sections of 4 um. Pretreated with 3% hydrogen peroxide for 20 min to quench endogenous peroxidases. Microwaved in antigen unmasking solution for 2-5 minutes as antigen retrieval.

Western Blot: NOD2 Antibody (2D9) [NB100-524] - Total protein from human THP1 and US-OS cells and mouse Raw264.7 cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-NOD2 in 1% non-fat milk in TBST and detected with an anti-mouse HRP secondary antibody using chemiluminescence.

Immunocytochemistry/Immunofluorescence: Doublecortin Antibody (3E1) [NBP1-92684] - Analysis of cortical neuron-glial cell culture from E20 rat stained with mouse mAb to Doublecortin (DCX), NBP1-92684, dilution 1:1,000 in red, and costained with chicken pAb to Microtubule Associated Protein2 (MAP2), dilution 1:10,000 in green. The blue is DAPI staining of nuclear DNA. DCX antibody reveals strong cytoplasmic staining in a population of small developing neurons and their processes, while MAP2 antibody stains dendrites and perikarya of mature neurons. Therefore DCX antibody is an excellent marker of developing neuronal cells.
Immunocytochemistry/Immunofluorescence: Doublecortin Antibody (3E1) [NBP1-92684] - Rat brain neural cultures stained with NBP1-92684 (green), a chicken polyclonal antibody to GFAP (NB300-213, red) and DNA (blue). The NBP1-92684 antibody reveals strong cytoplasmic staining in a population of small developing neurons. These cells are often found in small clumps as in these cultures, as in this example. Note that they are not positive for MAP2, which is characteristic of more mature neurons, and that the mature neuron shown is negative for NBP1-92684. This Doublecortin antibody is therefore an excellent marker of developing neuronal cells.
