Immunocytochemistry/Immunofluorescence: Caspase-6 Antibody (SC56-09) - (Pro and Active) [NBP2-67567] - Staining Caspase-6 in SW480 cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

Immunohistochemistry-Paraffin: Caspase-6 Antibody (SC56-09) - (Pro and Active) [NBP2-67567] - Analysis of paraffin-embedded human small intestine tissue with Rabbit anti-Caspase-6 antibody washed with ddH2O and PBS, and then probed with the primary antibody at 1/400 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

Caspases are a family of cytosolic aspartate-specific cysteine proteases involved in the initiation and execution of apoptosis. Caspase-6 is a major apoptotic executioner (1,2). It exists as a pro-domain protein and is cleaved by caspase-3 into two active subunits, an 18 kDa and a 10 kDa subunit (3). Caspase-6 cleaves among many different intracellular proteins, nuclear lamins. Lamins play a significant role in maintaining the integrity of the cell membrane, and their cleavage results in subsequent cellular breakdown and death (4).
