Background
Seizure Related Gene 6 (SEZ6), also known as Brain-specific Receptor-like Protein C (BSRP-C), is a member of the SEZ6 family of proteins with a predicted molecular weight of 150 kDa (1, 2). It is a type I transmembrane glycoprotein composed of an extracellular domain that contains three CUB domains and five short consensus repeats (SCRs) (1-3). Alternative splicing generates two additional isoforms for SEZ6. One isoform is a transmembrane protein that shows a deletion of amino acids (aa) 895-907 plus a 10 aa substitution for aa 982-991. The second splice variant is a soluble protein that shows an 18 aa substitution for aa 588-991 (1, 2). Mouse SEZ6 shares 91% and 97% aa sequence identity with the human and rat orthologs, respectively. SEZ6 is expressed in neurons in the developing and adult mouse brain and retina (1-6). It has been suggested that BSRP-C is important for proper development of dendritic arbors and excitatory synapses. SEZ6 has been shown to inhibit neurite branching of cortical neurons with little effect on neurite extension (7). Additionally, SEZ6 has been shown to interact with Motopsin/PRSS12, a serine protease believed to be essential for the reorganization of neuronal structures in the developing and adult brain (8). Mutations in
Sez6 have been associated with febrile seizures (9, 10).
