MKK3/MKK6 Antibody

Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line untreated (-) or exposed (+) to 100 J/m2of UV-C for 30 minutes and HepG2 human hepatocellular carcinoma cell line untreated (-) or treated (+) with 20 ng/mL Recombinant Human IL-1 beta /IL-1F2 (Catalog # 201-LB) for 20 minutes. PVDF membrane was probed with 5 µg/mL Rabbit Anti-Human Phospho-MKK3/MKK6 (MKK3 S218/T222, MKK6 S207/T211) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4930) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). Specific bands for Phospho-MKK3/MKK6 (MKK3 S218/T222, MKK6 S207/T211) were detected at approximately 40 and 38 kDa (as indicated). For additional reference, duplicate lysates were probed with 1 µg/mL Human/Mouse/Rat MKK3/MKK6 Monoclonal Antibody (lower panel, Catalog # MAB2514). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background

Mitogen-activated protein kinase kinase 3 (MKK3) and the closely related MKK6 are activated by proinflammatory cytokines and environmental stresses. Activation occurs through dual phosphorylation at S218/T222 of MKK3 and S207/T211 of MKK6 by several upstream MAP3Ks, including ASK1 and TAK1. Both MKK3 and MKK6 are dual-specificity protein kinases, phosphorylating and activating the p38 MAP kinases at T and Y positions within the phosphoacceptor sequence T-G-Y.
