Background

8D6 Antigen is a Type I membrane protein expressed by follicular dendritic cells. It shares significant homology to low density lipoprotein receptor (LDL-R) and has been shown to stimulate germinal center B cell growth.

Western blot shows lysates of HUVEC human umbilical vein endothelial cells and RPMI 8226 human multiple myeloma cell line. PVDF membrane was probed with 1 µg/mL of Sheep Anti-Human/Mouse/Rat Transcobalamin II Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7895) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Transcobalamin II at approximately 43 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Transcobalamin II was detected in immersion fixed paraffin-embedded sections of human kidney using Sheep Anti-Human/Mouse/Rat Transcobalamin II Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7895) at 10 µg/mL overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Sheep HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS019) and counterstained with hematoxylin (blue). DAPI reveals nuclei of neurons and glia in blue. The hippocampal neurons stain green for FOX3/NeuN and a few also are expressing c-FOS, and so appear orange. These cells were spontaneously active at the time the animal was sacrificed.

Then, after seven days of culture, 3x10^6 three-days PHA-activated human PBMC are treated with LAG-3 (human), mAb (17B4) (FITC) or FITC coupled isotype-matched (IgG1) control MAb (used at a saturating dilution of 1:800 and 1:150 respectively) for 30 min. at 4C in RPMI 1640 and washed twice with 1X PBS. Stained cells are then analysed by FC [4].
