AP3B1, also known as Adaptor protein 3 beta 3A, is specific for Adaptor Protein 3, one of four adaptor proteins interactive with endosomal Toll-like Receptors that mediate TLR trafficking and signaling cascades through the ER-endosomal systemIt has been shown specifically for TLR3, 7 and 9, all important innate immune anti-viral sensors, that interaction with UNC93B through AP-3 (AP3B1) allows loaded TLR transport from the endoplasmic reticulum to endolysosmes (1-4).The adaptor proteins AP-1, AP-2, AP-3, and AP-4 are utilized by UNC93B in trafficking and signaling pathways of TLRsThe AP3B1 antibody could be used to confirm AP-3 location on the endosomal membrane (4)AP-3 is critical for allowing TLR9 binding and delivery of its bound ligand, such as viral or parasitic RNA PAMP, from the ER to endolysomes which occurs normally in plasmacytoid dendritic cells.In AP-3 deficient cells, CpG -TLR9 activation of IFN expression does not occurThis dysregualtion is thought to be due to a failure of TLR9 engagement of IRF7 and TLR9 trafficking into the endolysosomeAP3B1 monitoring and AP3B1 quantitation therefore could serve as a model to examine the related TLR9 signaling, trafficking and activation pathway network.
