![Immunohistochemistry-Paraffin: Niemann-Pick C1 Antibody (1318A) [NBP2-76798] - Niemann-Pick Type C1/NPC1 was detected in immersion fixed paraffin-embedded sections of human brain (cerebellum) using Rabbit Anti-Human/Mouse/Rat Niemann-Pick Type C1/NPC1 Monoclonal Antibody (NBP2-76798) at 3 ug/mL for 1 hour at room temperature followed by incubation with the Anti-Rabbit IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC003). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). ![Immunocytochemistry/Immunofluorescence: Niemann-Pick C1 Antibody (1318A) [NBP2-76798] - HeLa cells were fixed and permeabilized for 10 minutes using -20C MeOH. The cells were incubated with anti-Niemann-Pick C1 Antibody (1318A) at 2 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). ![Immunohistochemistry-Paraffin: Presenilin-1 Antibody (APS 11) [NB300-749] - Immunohistochemistry was performed on normal biopsies of deparaffinized Human liver tissue. To expose target proteins, heat induced antigen retrieval was performed using 10mM sodium citrate (pH6.0) buffer, microwaved for 8-15 minutes. Following antigen retrieval tissues were blocked in 3% BSA-PBS for 30 minutes at room temperature. Tissues were then probed at a dilution of 1:200 with a mouse monoclonal antibody recognizing Presenilin 1 or without primary antibody (negative control) overnight at 4C in a humidified chamber. Tissues were washed extensively with PBST and endogenous peroxidase activity was quenched with a peroxidase suppressor.
