(B) Western blot results of SH-SY5Y treated with 100–500 nM celastrol show celastrol (500 nM) increased in protein expressions of Beclin 1 & Vps34 (phagophore induction), Atg7 (phagophore elongation), LC3-II (vacuole formation), DJ-1, & PINK1 (mitophagy). Celastrol (500 nM) suppressed LRRK2 expression. As control for the correct molecular weight, in vitro-transcribed & -translated (IVTT) recombinant survivin protein, derived from the full-length human cDNA was loaded. Furthermore, lysates from adult human cartilage served as a negative control. Total protein loaded was 1 μg for recombinant human survivin, 60 μg for chondrosarcoma & cartilage lysates. For A, B & E the polyclonal rabbit anti-survivin antibody AF886 was used. For C & D the monoclonal mouse anti-survivin antibody clone 32.1 was used.
