Background
CD200 R4, also known as CD200RLa, is a transmembrane protein in the immunoglobulin superfamily (1). Mature mouse CD200 R4 consists of a 216 amino acid (aa) extracellular domain (ECD), a 21 aa transmembrane segment, and an 8 aa cytoplasmic domain (2, 3). The ECD contains one Ig-like V-type domain and one Ig-like
C2-type domain. A positively charged lysine in the transmembrane likely mediates association with the signaling molecule DAP12 (2). CD200 R4 is expressed on monocytes, bone marrow derived macrophages and dendritic cells, NK cells, and resting mast cells. It is down-regulated following Fc epsilon RI-mediated mast cell activation (2). According to conflicting reports, CD200 R4 may or may not interact with CD200 (3, 4). R&D Systems in-house testing indicates that this recombinant mouse CD200 R4 binds to recombinant mouse CD200-Fc.
