Flow Cytometry: p53 Antibody (PAb 240) [NB200-103] - An intracellular stain was performed on HeLa cells with p53 Antibody [PAb 240] NB200-103 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1.0 ug/mL for 30 minutes at room temperature, followed by Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (35503, Thermo Fisher).

Immunocytochemistry/Immunofluorescence: p53 Antibody (PAb 240) [NB200-103] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-p53 (PAb 240) [NB200-103] at a 1:200 dilution overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunocytochemistry/Immunofluorescence: p53 Antibody (PAb 240) [NB200-103] - HeLa cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-p53 Antibody (PAb 240) NB200-103 at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Flow Cytometry: p53 Antibody [NB200-171] - p53 in Jurkat Cells, fixed in 1.5% PFA, and permeabilized in 90% Methanol. 1 million cells were stained with 1.5 ug anti-KLH IgG control or anti-p53 NB200-171 and secondary FITC-conjugated goat anti-rabbit (in a 150ul reaction). Black- anti-KLH control IgG; Red- anti-HDMX.

Chromatin Immunoprecipitation Sequencing: p53 Antibody [NB200-171] - Chromatin from MCF7 was immunoprecipitated with anti-p53 antibody and analyzed by DNA sequencing. The figure illustrates the peak distribution of p53 binding within a 250 Kb region of chromosome 6 as detected using anti-p53. ChIP-seq validation performed by Active Motif, Carlsbad, CA.

Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HEK293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-P53 antibody NB200-171 used for IP at 3 ug per reaction. P53 was also immunoprecipitated by rabbit anti-P53 antibody NB100-2204. For blotting immunoprecipitated P53, NB200-171 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Immunohistochemistry-Paraffin: p53 Antibody (5E2) [NBP2-00723] - Staining of paraffin-embedded Carcinoma of Human pancreas tissue using anti-p53 mouse monoclonal antibody.

Flow Cytometry: p53 Antibody (5E2) [NBP2-00723] - HEK293T cells transfected with either overexpression plasmid (Red) or empty vector control plasmid (Blue) were immunostained by anti-p53 antibody, and then analyzed by flow cytometry.

Western Blot: p53 Antibody (5E2) [NBP2-00723] - HEK293T cells were transfected with the pCMV6-ENTRY control (Left lane) or pCMV6-ENTRY p53 (Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-p53.

<b>Biological Strategies Validation. </b>Immunoprecipitation: p53 Antibody [NBP2-19667] - Immunoprecipitation of p53 protein. HCT116 lysates with 30uM cisplatin treatment for 24 hours were subjected to immunoprecipitation using (B) normal rabbit IgG or (C) 2. 5 ug of anti-p53 antibody. (A) Input, 20ug of HCT116 lysates. The precipitated protein was detected by NBP2-19667. anti-Rabbit IgG Kit was used in the Western blot.

Whole cell extract (30 ug) were separated by 10% SDS-PAGE, and the membrane was blotted with p53 antibody [N1], N-term (NBP2-19667) diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.
