The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. [1] protein standard, [2] rat brain, [3] mouse brain, [4] NIH-3T3, [5] HeLa, [6] HEK293, and [7] C6 cells. The DJ1 antibody detects protein with apparent molecular weight of 21kDa but only in human cell lines, since it does not recognize the rat or mouse DJ1 protein.
