![<b>Genetic Strategies Validation. </b>Immunohistochemistry-Paraffin: Neurofibromin 1 Antibody (McNFn27a) [NB300-153] - IHC analysis of formalin fixed paraffin-embedded (FFPE) human brain using Neurofibromin 1 antibody at 1:50 on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Nuclear staining of Neurofibromin 1was observed. [1] protein standard, [2] rat brain, [3] mouse brain, [4] NIH-3T3, [5] HeLa, [6] HEK293, and [7] C6 cells. The DJ1 antibody detects protein with apparent molecular weight of 21kDa but only in human cell lines, since it does not recognize the rat or mouse DJ1 protein. ![Immunohistochemistry Free-Floating: NF-M Antibody (3H11) [NB300-134] - Adult rat frontal cortex section stained with mouse mAb to neurofilament NF-M, dilution 1:5,000 in green, and costained with chicken pAb to neurofilament NF-H, dilution 1:5,000 in red. Following transcardial perfusion of rat with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45uM, and free-floating sections were stained with above antibodies.
