Temporally coordinated expression of Sox2, Tbr2, NueroD, and Prox1 in gfap-GFP+ GNPs and differentiating granule neurons Gfap-GFP+/Sox2+ progenitor cells were found at the dentate neuroepithelium at E14 (arrow in A1). Sox2 expression was found at the VZ. Tbr2+ GNPs are then observed (red, arrow in A2). Tbr2 expression is followed by NeuroD expression (A3). Tbr2+/NeuroD+ GNPs are found (arrow, A3). The Tbr2+ GNPs (red) migrate toward the DG primordium and co-express Prox1 (green), a marker for early differentiating dentate granule neurons (arrow in A4). Gfap-GFP+ progenitors sequentially express Sox2, Tbr2, NeuroD1, and then, Prox1 in the developing DG at E16 (arrows in B1–B4, respectively). Neural induction of hPSCs. (a) Both hPSC culture and neural differentiation were performed on LN521 substrate. Cortical neurons were induced with dual SMAD inhibition, expanded in the presence of FGF2, and matured with support by a selection of neurotrophic factors. Neural progenitor cells (NPCs) could be cryopreserved at day 21 and plated at day 32 for final experiments, including microelectrode array (MEA) measurements. (b) Three different hPSC lines (one hESC line, 08/023 and two hiPSC lines, 10212.EURCCs and IMR90-4) were characterized for their efficiency in producing neuroectodermal cells in response to 12-day neural induction by dual SMAD inhibition. Cells were stained for pluripotency marker Oct4 at the pluripotent stage and after 12 days of neural induction. The presence of early neuroectodermal markers was evaluated with Sox2, FoxG1 and Pax6 staining. (c) After 21 days of differentiation, the culture contained vimentin- and Pax6-positive NPCs that could be cryopreserved. Additionally, the first Tbr2- and MAP2-positive neurons were detected at this time point. (d) The percentages of Pax6-positive cells were quantified at four time points of differentiation (mean ± s.e.m., n = 5–14, data derived from 1–3 independent differentiations). Immunohistochemistry: SOX2 Antibody (PO00-28) [NBP3-32972] - Immunohistochemical analysis of paraffin-embedded human trachea tissue with Rabbit anti-SOX2 antibody (NBP3-32972) at 1/1,000 dilution.<br/><br/> The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32972) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Immunocytochemistry/ Immunofluorescence: SOX2 Antibody (PO00-28) [NBP3-32972] - Immunocytochemistry analysis of F9 cells labeling SOX2 with Rabbit anti-SOX2 antibody (NBP3-32972) at 1/250 dilution.<br/><br/> Cells were fixed in 4% paraformaldehyde for 20 minutes at room temperature, permeabilized with 0.1% Triton X-100 in PBS for 5 minutes at room temperature, then blocked with 1% BSA in 10% negative goat serum for 1 hour at room temperature. Cells were then incubated with Rabbit anti-SOX2 antibody (NBP3-32972) at 1/250 dilution in 1% BSA in PBST overnight at 4 ℃. Goat Anti-Rabbit IgG H&L (iFluor™ 488) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. Nuclear DNA was labelled in blue with DAPI.<br/><br/> Beta tubulin (red) was stained at 1/100 dilution overnight at +4℃. Goat Anti-Mouse IgG H&L (iFluor™ 594) was used as the secondary antibody at 1/1,000 dilution.

Western blot analysis on different lysates with NBP3-32972 at 1/2,000 dilution. Lane 1: NCCIT cell lysate Lane 2: F9 cell lysate Lane 3: HeLa cell lysate (negative) Predicted band size: 34 kDa Observed band size: 36 kDa Exposure time: 5 seconds; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. NBP3-32972 at 1/2,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG [HRP] Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Western blot analysis on different lysates with NBP3-32972 at 1/1,000 dilution. Lane 1: Mouse brain tissue lysate Lane 2: Rat brain tissue lysate Lysates/proteins at 30 µg/Lane. Predicted band size: 34 kDa Observed band size: 36 kDa Exposure time: 25 seconds; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. NBP3-32972 at 1/1,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG [HRP] Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.
