Immunocytochemistry/Immunofluorescence: c-Myc Antibody (9E10) [NB600-302] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with anti-c-Myc (9E10) at 10 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Flow (Intracellular): c-Myc Antibody (9E10) [NB600-302] - An intracellular stain was performed on U-937 cells with c-Myc Antibody (9E10) NB600-302PE (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Phycoerythrin.. Using the PE format of this antibody.

Flow Cytometry: c-Myc Antibody (9E10) [NB600-302] - An intracellular stain was performed on U-937 cells with c-Myc Antibody (9E10) NB600-302AF488 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 488.

Western Blot: c-Myc Antibody (9E10) - BSA Free [NB600-302] - NLRP2 interacts with SCMC components TLE6, OOEP, FILIA & NLRP5.(A) NLRP2 was overexpressed with myc-tagged OOEP, TLE6, NLRP5 & FILIA in HEK293T cells for 48 hours, immunoprecipitated with anti-NLRP2 or IgG as negative control & immunoblotted with anti-myc. Top panel shows specificity of anti-NLRP2 IP & bottom panel shows that NLRP2 binds to OOEP, TLE6, FILIA & NLRP5. Uncropped, full length western blots have been provided in Supplementary Figure 5. (B) Whole-mount immunofluorescence co-staining with anti-NLRP2 (green) & DAPI (blue) for nuclear staining on Nlrp2+/+ oocytes & embryos at 2-, 4-,16-cell & morula stages revealed a predominantly SCMC-like localization for NLRP2. Western Blot: c-Myc Antibody [NB600-336] - Analysis of whole cell lysate (WCL) of Jurkat cells using anti-cMyc antibody (NB600-336) at 1:1000 dilution. HRP conjugated goat anti-rabbit IgG (H+L) cross adsorbed secondary antibody was used with ECL substrate for the detection of c-Myc antibody bound to the blotted protein. This c-Myc antibody detected the c-Myc specific band at its expected position (48-50kDa). The signal below 10 kDa in this blot is potentially the degraded protein and we have not characterized this band.

Our data indicated that upon acquired resistance in HNSCC cells or in non-responder HNSCC tumors, the levels of miR124-3p and miR766-3p go up, which in turn down-regulate its direct target genes: CREBRF and NR3C2, and consequently the expression of downstream targets of CREBRF (ATG5/CREB3) and NR3C2 ( beta -catenin/c-Myc) increased in resistant tumors, which are positively correlated with poor prognosis. Thus, by enhancing the CREBRF-ATG5/CREB3 and NR3C2-beta -catenin/c-Myc axis, miR124-3p and miR766-3p support aggressive HNSCC progression. Flow Cytometry: c-Myc Antibody (9E11) [NB200-108] - An intracellular stain was performed on U937 cells with c-Myc [9E11] Antibody NB200-108AF594 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 594.

Western Blot: c-Myc Antibody (9E11) [NB200-108] - Whole cell protein from PC3 (lane 1), U-2 OS (lane 2) and mouse testis (lane 3) was separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-c-Myc in 1% milk, and detected with an anti-mouse HRP secondary antibody using chemiluminescence.

Flow Cytometry: c-Myc Antibody (Myc.A7) [NBP2-37822] - An intracellular stain was performed on Jurkat cells with c-Myc Antibody (Myc.A7) NBP2-37822AF488 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 488.

Western Blot: c-Myc Antibody (Myc.A7) [NBP2-37822] - Analysis using the DyLight 680 conjugate of NBP2-37822. Detection of Myc Epitope Tag was performed by loading various amounts of E. coli lysate containing a multi-epitope tagged protein per well onto a 4-20% Tris-HCl polyacrylamide gel. Proteins were transferred to a PVDF membrane and blocked with 5% BSA/TBST for at least 1 hour. The membrane was probed with a DyLight 680-conjugated Myc Epitope Tag monoclonal antibody at a dilution of 1:1000 for 1 hour at room temperature on a rocking platform and washed in TBS-0.1% Tween-20. Detection was performed using the LI-COR Odyssey.
