Axl phosphorylated at Y779 (panels B, D) and total Axl (panels A, C) were assessed in immersion fixed A172 human glioblastoma cells incubated with (panels C, D) or without (panels A, B) pervanadate. Phospho-Axl was detected using Rabbit Anti-Human Phospho-Axl (Y779) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2228) at 10 µg/mL for 3 hours at room temperature. Western blot shows lysates of A172 human glioblastoma cell line untreated (-) or treated (+) with 1 mM Pervanadate (PV) for 30 minutes. PVDF membrane was probed with 0.2 µg/mL of Mouse Anti-Human Phospho-Axl (Y779) Monoclonal Antibody (Catalog # MAB6965) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for Phospho-Axl (Y779) at approximately 140 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

<b>Biological Strategies Validation. </b>Axl phosphorylated at Y779 and total Axl were assessed in immersion fixed A172 human glioblastoma cells incubated with (upper panel) or without (lower panel) the phosphatase inhibitor pervanadate at 100 µM for 5 minutes. Phospho-Axl was detected using Mouse Anti-Human Phospho-Axl (Y779) Monoclonal Antibody (Catalog # MAB6965) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained using DAPI (blue). Total Axl was detected using Goat Anti-Human Axl Antigen Affinity-purified Polyclonal Antibody (Catalog # AF154). Cells were stained using the NorthernLights™ 493-conjugated Anti-Goat IgG Secondary Antibody (green; Catalog # NL003). Specific staining was localized to cytoplasm and cell surfaces. <b>Biological Strategies Validation. </b>Simple Western lane view shows lysates of A172 human glioblastoma cell line untreated (-) or treated (+) with 1 mM Pervanadate (PV) for 30 minutes, loaded at 0.2 mg/mL. A specific band was detected for Phospho-Axl (Y779) at approximately 150 kDa (as indicated) using 4 µg/mL of Mouse Anti-Human Phospho-Axl (Y779) Monoclonal Antibody (Catalog # MAB6965). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.*Non-specific interaction with the 230 kDa Simple Western standard may be seen with this antibody.

Human glioblastoma cell line, A172, was treated with conditioned media from the human lung cell line, WI38, for fifteen minutes to induce tyrosine phosphorylation of Axl. Serial dilutions of lysates were analyzed by (A) IP-Western blot and (B) this ELISA . IPs were done using an anti-Axl monoclonal antibody and anti-mouse IgG agarose. Immunoblots were incubated with a biotinylated anti-phosphotyrosine monoclonal antibody (Catalog # BAM1676) to detect phospho-Axl (p-Axl). Bands were visualized with Streptavidin-HRP (Cat # DY998) followed by chemiluminescent detection.

<b>Biological Strategies Validation. </b>A172 cells were untreated or treated with conditioned media from the human lung cell line, WI38, for fifteen minutes. ELISA and IP-Western blot (inset) analyses were done using 100 μg and 400 μg of lysate, respectively. IP-Western blots for phospho-Axl (p-Axl) were done as described in Figure 1. Blots were stripped and total Axl was detected using a biotinylated anti-Axl polyclonal antibody (Catalog # BAF154).

<b>Biological Strategies Validation. </b>A172 cells were treated with conditioned media from the human lung cell line WI38 for fifteen minutes. The indicated amounts of recombinant extracellular domains of human Axl (Catalog # 154-AL), human Dtk (Catalog # 859-DK) or human Trk A (Catalog # 175-TK) were added to 125 μg lysate and analyzed using this ELISA. Competition was observed only with recombinant Axl.

Lysates prepared from the human glioblastoma cell line A172 were diluted in series and analyzed by (A) IP-Western Blot and (B) this DuoSet® IC ELISA. IPs were performed using an anti-Axl monoclonal antibody and goat anti-mouse agarose. Immunoblots were incubated with a Biotinylated Anti-Axl Polyclonal Antibody (Catalog # BAF154) to detect human total Axl. Bands were visualized with Streptavidin-HRP A (Catalog # DY998) followed by chemiluminescent detection. Human Axl can be detected in this DuoSet® IC ELISA by using approximately 10 to 20 times less lysate than is needed for a conventional IP-Western Blot.

Lysates were prepared from the human glioblastoma cell line (A172), the human epidermoid carcinoma cell line (A431), and the human stomach cancer cell line (KatoIII). DuoSet® IC ELISA and IP-Western Blot (inset) analyses were performed using 50 μg and 100 μg of lysate, respectively. The IP-Western Blot was performed as described in Figure 1. The DuoSet® IC ELISA results correlate well with the amounts of human Axl detected by IP-Western Blot.
