302-B2) was serially diluted and captured by Mouse Anti-TGF‑ beta 2 Monoclonal Antibody (Catalog # MAB612) coated on a Clear Polystyrene Microplate (Catalog # DY990). Goat Anti-TGF‑ beta 2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-302-NA) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (Catalog # DY998)" class="big_lightbox" target="_blank">
HAF017). A specific band was detected for TGF-beta 2 at approximately 70 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1." class="big_lightbox" target="_blank">

102-B2) inhibits Recombinant Mouse IL-4 (Catalog # 404-ML) induced proliferation in the HT-2 mouse T cell line in a dose-dependent manner (orange line). Inhibition of Recombinant Mouse IL-4 (7.5 ng/mL) activity elicited by Porcine TGF-beta 2 (1 ng/mL) is neutralized (green line) by increasing concentrations of Sheep Anti-TGF-beta 2 Polyclonal Antibody (Catalog # AB-112-NA). The ND50 is typically 0.3-1.5 µg/mL." class="big_lightbox" target="_blank">

341-BR) was captured on a Sensor Chip Protein AGL, and binding to Recombinant Human TGF-beta 2 (302-B2) was measured at a concentration range between 0.0488 nM and 100 nM. The double-referenced sensorgram was fit to a two-state reaction binding model to determine the binding kinetics and affinity, with an affinity constant of KD=22.83 nM. Transforming Growth Factor (TGF) betas mediate many cell to cell interactions that occur during embryonic development. Three TGF betas have been identified in mammals. TGF beta 1, TGF beta 2 and TGF beta 3 are each synthesized as precursor proteins that are very similar in that each is cleaved to yield a 112 amino acid polypeptide that remains associated with the latent portion of the molecule. The TGF beta polypeptides are multifunctional; capable of influencing cell proliferation, differentiation, and other functions in a wide range of cell types. Transformed, as well as nonneoplastic tissues, release transforming growth factors; and essentially all mammalian cells possess a specific TGF receptor. The multi modal nature of TGF beta is seen in its ability to stimulate or inhibit cellular proliferation. In general, cells of mesenchymal origin appear to be stimulated by TGF beta whereas cells of epithelial or neuroectodermal origin are inhibited by the peptide. TGF beta 1, TGF beta 2, and TGF beta 1.2 appear to be equivalent in biological activity, although there does appear to be differences in binding to certain types of receptors. TGF beta 2 is produced by many cell types and has been found in the highest concentration in porcine platelets and mammalian bone. Latent TGF beta 2 is the prominent isoform found in body fluids such as amniotic fluid, breast milk, and the aqueous and vitreous humor of the eye.
