Enzyme-linked immunosorbent assay (ELISA) is an analytic biochemical assay that utilizes enzyme-linked antibodies on a microwell platform to quantitate specific proteins within biological samples. Visual detection is then achieved by adding the enzyme’s substrate, which causes a color-changing reaction to occur. There are three different types of ELISA that are most commonly used: indirect ELISA, sandwich ELISA and competitive ELISA. These assays are extremely useful for high-throughput diagnostic screenings drug testing.

Following antigen retrieval, tissues were blocked in 3% H2O2-methanol for 15 min at room temperature, washed with ddH2O and PBS, and then probed with a Beta-Tubulin loading control antibody diluted in 3% BSA-PBS at a dilution of 1:20 overnight at 4C in a humidified chamber. Tissues were washed extensively in PBST and detection was performed using an HRP-conjugated secondary antibody followed by colorimetric detection using a DAB kit.
