![Immunoprecipitation: Dynamin Antibody (3G4B6) [NB110-60491] - Antibody-bead conjugates were prepared by adding 1.0 ug of antibody to 500 ul of PBS with 0,01% triton X-100 in a microcentrifuge tube and 30ul of protein G Sepharose beads. Tubes were rocked O/N at 4 degrees C followed by several washes to remove unbound antibodies. U2OS were collected in HEPES buffer (20 mM HEPES, 100 mM sodium chloride, 1 mM EDTA, 1% Triton X-100, pH 7.4) supplemented with protease inhibitor. Lysates are rocked 30 min at 4 degrees C and spun at 110,000xg for 15 min at 4 degrees C. One ml aliquots at 1 mg/ml of lysate were incubated with an antibody-bead conjugate for ~2 hrs at 4 degrees C. Following centrifugation, the unbound fractions were collected, and beads were subsequently washed three times with 1 ml of HEPES lysis buffer and processed for SDS-PAGE and immunoblot. ![Immunohistochemistry: Dynamin Antibody (A1B2-R) [NBP3-32283] - Immunohistochemical analysis of paraffin-embedded human brain tissue with Mouse anti-Dynamin antibody (NBP3-32283) at 1/1,000 dilution. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32283) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen.
