![Chromatin Immunoprecipitation: CREB Antibody [NBP2-22516] - Analysis performed using cross-linked chromatin from colon carcinoma cells treated with serum. IP was performed using a multiplex microplate Matrix ChIP assay of a CREB polyclonal antibody. Chromatin aliquots from cells were used per ChIP pull-down. Quantitative PCR data one in quadruplicate using 1ul of eluted DNA in 2ul SYBR real-time PCR reactions containing primers to amplify -15kb upstream of Egr1 or exon-1 or exon-2 of Egr1.Quantitation of immunoprecipitated chromatin is presented as signal relative to the total amount of input chromatin. (A) Whole cell lysates of MCF-7 & MDA-MB-231 were prepared after 6 hours of exposure to DCQ (5 μM) under normoxia or hypoxia, & blots were probed for p-H2AX, p53, p-p53, p21, HIF-1 alpha & GAPDH. Results are representative of three independent experiments. (B) MDA-MB-231 & MCF-7 cells were pretreated with 1 mM Vitamin E or DTT for 2 hours followed by 25 min incubation with 10 μM CM-H2DCFDA dye. Cells were washed with PBS & treated with DCQ for 1 hour under normoxia or hypoxia, after which cells were harvested & the amount of DCF fluorescence was analyzed by flow cytometry. Each percentage is the average ± SE of three independent experiments.
