A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected band size for alpha Tubulin is ~55kDa. ![Immunohistochemistry-Paraffin: alpha Tubulin Antibody (F2C) - Chimeric [NBP3-09006] - Anti-Alpha Tubulin staining of paraffin embedded human cerebral cortex tissue using the rabbit-chimeric version of F2C (NBP3-09006). Antigen retreival was acheived by microwaving in citrate buffer (pH6), followed by blocking with protein block serum-free buffer. Primary antibody incubation with NBP3-09006 was carried out at 4 ug/ml for 30 minutes. Samples were then incubated with an anti-rabbit IgG HRP secondary antibody for 20 mins followed by DAB (3,3'-diaminobenzidine), and counter-staining with haemotoxylin. Staining of axons and a few neuronal cell bodies may be observed. ![<b>Independent Antibodies Validation. </b>Western Blot: BIN1 Antibody [NBP1-46170] - Whole cell lysate (5, 15 and 50 mcg for WB; 1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-Bin1 antibody used for WB at 0.1 mcg/ml (A) and 1 mcg/ml (B) and used for IP at 3 mcg/mg lysate. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-Bin1 antibody NBP1-46170 used for IP at 6 ug per reaction. Bin1 was also immunoprecipitated by another rabbit anti-Bin1 antibody. For blotting immunoprecipitated Bin1, NBP1-46170 was used at 1 ug/ml.
