Prolyl-hydroxylase Domain Containing Protein 2 (PHD2) is one of four hydroxylase enzymes that function as oxygen sensors. They are responsible for the post-translational modification of hypoxia-inducible factor 1 alpha (HIF-1 alpha), a component of the transcriptional complex involved in oxygen homeostasis. PHDs catalyze the hydroxylation of prolyl residues on HIF-1 alpha and target it for proteasomal degradation via von Hippel-Lindau ubiquitination. The four isoforms appear to function in a non-redundant manner and appear to differ in their expression patterns and catalytic selectivity. PHD2 antibody provided new insights into the complex formation partners of the melanoma antigen-11 (MAGE-11) protein in a yeast two-hybrid screen (1).
Novel signaling pathways for transforming growth factor beta 1 (TGB1) in fibrosis and chronic kidney disease states were found to involve PHD2 via a SMAD2/SMAD3 inhibitor-dependent pathway in studies with PHD2 antibody (2). Cancer cell survival and cell cycle progression have also been followed using the PHD2 antibody to demonstrate that its related family member PHD3 enhances the hypoxic survival of tumor cells in a head and neck squamous cell carcinoma (HNSCC) model (3) while further studies with PHD2 antibody in a similar HNSCC system link the overexpression of PHD2 protein with key metrics such as tumor aggressiveness, less differentiated phenotypes, and elevated proliferation levels (4). Another study used the PHD2 antibody to characterize the role of presenilin (PSEN) in oxygen-sensing in Alzheimer’s disease pathogenesis by studying a variety of genetically modified cell and mouse lines (5).
