Proliferating cell nuclear antigen (PCNA) is an evolutionarily well-conserved protein found in all eukaryotic species as well as in Archaea. PCNA was first shown to be involved in DNA replication. However PCNA functions are associated with other vital cellular processes such as chromatin remodeling, DNA repair, sister-chromatid cohesion and cell cycle control as well (1). PCNA is considered to be a marker of cell proliferation in various cancers. Immunohistochemical staining using PCNA antibody was performed on skin from patients with various malignant and nonmalignant skin diseases.

The percentage of PCNA-positive cells was highest for the malignant diseases than those for the normal control or nonlesional skin of the patients. The results suggest that the extent of PCNA antibody staining in human skin diseases, which is a cofactor of DNA polymerase-delta and is essential for cell proliferation, correlates with the extent to which the disease is treatment-resistant. Similar observations on the role of PCNA in various cancers suggest that distribution of PCNA-positive cells may be helpful in the early diagnosis of cancers (2, 3). Determining the pattern of PCNA-positive cells is easy and does not require cell counting, thereby reducing the need to use the radioactivity in cell counts (4).
