Survivin is an anti-apoptotic protein from a large family with related members such as X-linked IAP, cIAP1 and cIAP2, IAP-like protein-2, melanoma IAP, Livin, and NAIP. Survivin regulates fundamental physiological events including the cell cycle, fetal development, and cell migration. To further understand apoptotic pathway mechanisms, Xu’s group at MD Anderson used the survivin antibody in their DNA microarray screen of promyelocytic leukemia protein (PML) target genes (1). There, they were able to identify a novel PML4-dependent pathway with survivin as a main downstream target.

Survivin antibody enabled identification of a key role of the PI3-kinase/Akt/NFkB/survivin signal in an HCAEC culturing system by using hypoxic preconditioning to enhance tubule formation and accelerated morphogenesis (2). In studies looking investigating new anticancer therapies for highly invasive urinary bladder cancer (iUC), survivin antibody was again used in microarray screens that eventually identified DNA methyltransferase 1 (DNMT1) as a strong candidate for antiproliferative inhibitors in both canine and human iUC models (3). The role of survivin in another cancer type – that of breast cancer – was also studied using the survivin antibody in the Ryan lab (4). This particular group compared the expression levels of various survivin splice isoforms in normal versus adenomas and primary carcinomas and found different correlations between particular isoforms and disease types. Furthermore, colon cancer studies using survivin antibody immunhistochemistry demonstrated different subcellular localization patterns for wild-type and mutant forms of the adenomatous polyposis coli protein APC tumor suppressor, with interesting insights into how particular APC mutations govern tumor cell survival (5).
