Background
Tetracycline repressor TetR (tetracycline repressor protein class B from transposon Tn10) belongs to the Tet/Cam family of bacterial regulators and it controls the expression of Tn10 encoded tetracycline resistance. When tetracycline is not present, TetR recognises its operator sequence, tetO, for repressing tetA expression (resistance gene) and its own expression. Upon entry into the cell, tetracycline binds a pocket within the protein core of TetR and induces a conformational change leading to reorientation of DNA binding heads, which in turn abolishes operator binding for triggering resistance gene expression. TetR repressed tetracycline resistance element is responsible for the maintenance of antibiotic resistance traits in strains of Escherichia coli which confers an advantage in selection to E. coli expressing tetracycline resistance. Moreover, TetR has attracted widespread interest because regulators belonging to this family control the expression of genes which are involved in development, fatty acid metabolism, drug resistance and many other cellular processes.
