Penicillinase Antibody Summary

This antibody is purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer stated above.
Penicillinase (Enterobacter cloacae).
Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum as well as purified and partially purified Penicillinase [E.clocae]. Cross reactivity against Penicillinase from other tissues and species may occur but have not been specifically determined.
Ion exchange chromatography

Penicillinase is a beta-lactamase, showing specificity for penicillins by hydrolysing the beta-lactam ring. Beta lactamases play a significant role in the high resistance of certain organinsms to penicillins, cephalosporins, cephamycins and carbapenems. These antibiotics have a common element in their molecular structure: a four-atom ring known as a beta-lactam. The lactamase enzyme breaks that ring open, deactivating the molecule's antibacterial properties. Penicillinase resistant beta lactams have been developed, such as methicillin, although there is widespread resistance to these now also (MRSA).
