Background
Nsp1p represents one of the most abundant nucleoporins (Nups) and is the only Nups member known to form two distinct NPC subcomplexes which are (i) Nic96p: this complex is composed of Nsp1p, Nup57p, Nup49p, and Nic96p, and is localized to both sides of the central gated channel and to the nuclear basket (ii) Nup82p: this complex is consists of Nsp1p, Nup82p, and Nup159p and is found exclusively on the cytoplasmic phase of nuclear pore complex. Moreover, Nup82p complex has also been documented to associate with Nup116p-Gle2p subcomplex. Both of these Nsp1p complexes (Nic96p and Nup82p) are implicated in delivering critical roles in nucleo-cytoplasmic transport. Genetic mutation or induced depletion of Nsp1p as well as of Nic96p, Nup57p, or Nup49p results in defective protein import. Nup82 or Nup159 mutants depict an expediated onset of nuclear mRNA retention with no impact on nuclear protein import. Moreover, Nsp1p, Nup49p and Nic96p appear to implicate in export of ribosomal large subunits, whereas Nup49 mutants are impaired in mRNA export.
