Background

FUNCTION: Guanine nucleotide-binding proteins (G proteins) are involved as a modulator or transducer in various transmembrane signaling systems. The beta and gamma chains are required for the GTPase activity, for replacement of GDP by GTP, and for G protein-effector interaction. SUBUNIT: G proteins are composed of 3 units, alpha, beta and gamma. Component of the RGS9-1-Gbeta5 complex composed of RGS9 (isoform RGS9-1), Gbeta5 (GNB5) and RGS9BP. TISSUE SPECIFICITY: Expressed in multiple tissues.

1. I was attempting to perform IHC on retinal tissue sections using the GBB5 antibody. May I have more information on the protocol used to do the IHC in the shown Human Kidney sections?
   • We did not use a customized protocol for this antibody. Please see our general protocols. 1. Antigen retrieval was performed using 0.01 M sodium citrate buffer, pH 6.0 at 95-100C for 20 minutes followed by slow cooling achieved through letting slides stay in buffer at room temperature for about 25-30 minutes. 2. Our lab used a working concentration of 20 ug/ml of primary antibody and the incubation time was about 1 hour at room temperature. However, the incubation time may vary for your samples and it depends upon several factors such as the extent of target expression in the tissues, tissue processing, epitope retrieval etc. 3. We used anti-Rabbit IgG antibody Biotin conjugated and depending upon your for detection system employed, you may choose a relevant secondary antibody from our catalog.
