ER associated Degradation Antibody Pack

Knockdown Validated: ER associated Degradation Antibody Pack [NBP1-78944] - Contribution and interaction of OS-9 for degradation of SHH proteins. Cells stably expressing the N278A-HA precursor were depleted of OS-9 by siRNA, and the fate of N278A was followed after cycloheximide (CHX) addition. All samples were analyzed by SDS-PAGE and immunoblotting with HA antibodies. Controls were treated with a commercial negative control siRNA. The samples were also immunoblotted for OS-9 to show the extent of depletion. Immunoblotting with GAPDH antibodies served as loading control. Western Blot: ER associated Degradation Antibody Pack [NBP1-78944] - Detection of Derlin protein in canine KRM microsomes using NB100-448

Immunohistochemistry: ER associated Degradation Antibody Pack [NBP1-78944] - FFPE tissue section of human tonsil using rabbit polyclonal HRD1 antibody (NB100-2526) at 2 ug/mL concentration (1:500 dilution). The primary antibody binding to SYVN1/HRD1 antigen in cells was detected using HRP conjugated anti-Rabbit secondary antibody with DAB reagent, and the sections were further counterstained with hematoxylin for labeling cellular nuclei. This HRD1 antibody generated an expected cytoplasmic staining of HRD1 with stronger signal around periphery of the nucleus signifying the staining of the endoplasmic reticulum.

Background

Protein folding is monitored by multiple, sometimes overlapping, surveillance mechanisms within the ER. When erroneous protein folding is detected, the proteins are recycled through the ER associated degradation pathway. OS9 and HRD1 coordinate to induce protein degradation in this pathway; whereas, through an independent mechanism Derlin1 also affects ER associated degradation. Analysis of these proteins can assist in identifying the point of dysfunction when ER associated degradation is ineffective.
