HES-4 was detected in immersion fixed HEK293 human embryonic kidney cell line using Sheep Anti-Human HES-4 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3600) at 1.7 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Sheep IgG Secondary Antibody (red; Catalog # NL010) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. Western blot shows lysates of PC-3 human prostate cancer cell line. Gels were loaded with 30 µg of whole cell lysate (WCL), 20 µg of cytoplasmic (Cyto), and 10 µg of nuclear extracts (Nuc). PVDF membrane was probed with 1 µg/mL Mouse Anti-Human HES-4 Monoclonal Antibody (Catalog # MAB3600) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band for HES-4 was detected at approximately 32 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 4.
